Evaluation of anti-thrombotic efficacy of newly developed catheter coated with urokinase together with heparin

We developed an antithrombotic catheter coated with both urokinase and heparin on its surface and evaluated its performance in vitro. Using a modified Chandler Loop method with human whole blood, the catheter demonstrated higher antithrombotic properties compared to those coated with urokinase or heparin alone, and these properties were maintained even after a 25-day rinse period. TAT and D-dimer levels in the blood within the loop increased with the extended rinse period, with a significant rise in D-dimer levels following the increase in TAT. Measurements of urokinase and heparin activity on the catheter surface, using synthetic substrates S-2251 and S-2238 for plasmin and thrombin, respectively, confirmed that both activities were retained even after the 25-day rinse period. It was revealed that coating the catheter surface with both heparin and urokinase resulted in the manifestation of two distinct antithrombotic actions—anticoagulant and fibrinolytic activation—thus maintaining long-term antithrombotic properties.
